Microsoft says more than 10,000 retail locations in over 40 countries will open at 12:01am on November 6 to sell Halo 4.
There will be more than 350 midnight store openings across the UK, including 292 GAME stores.
The flagship London launch will take place at GAME in Westfield Stratford, where fans will have the opportunity to meet developers from 343 Industries, get early hands-on time with the game, and win Halo prizes.
More than 30 global launch events are planned in major cities worldwide including London, Seattle, Paris, Berlin, Toronto, Singapore, Tokyo, Hong Kong, Dubai and Sydney.
London will host "a spectacular stunt" on the evening of Monday November 5, down the River Thames from the Greenwich Peninsula to Tower Bridge. Fans are advised to arrive at Butlers Wharf, adjacent to the Design Museum, no later than 7:30pm.
